LOCAL PRODUCTION AND STANDARDIZATION OF ANTI-HUMAN GLOBULIN (AHG)

ABSTRACT

Anti-human globulin is a reagent used in the detection of incomplete antibodies. The purpose of this study is to know if AHG can be produced locally. The study was carried out using 2 rabbits which were 16 weeks old each and Dunsfords and Bowley’s method of AHG production was used. The rabbits were immunized with human O serum mixed with a freund’s adjuvant Phosphate buffered saline. The locally prepared AHG was actually a working reagent with a liter of 1\32 and when it was used in parallel with commercially prepared AHG for direct and indirect coomb’s tests, the results generated were as expected. In conclusion, the local preparation of AHG should be encouraged due to the fact that it would really be a beneficial venture by bringing out the zeal in us as scientist to prepare our reagent locally not to depend only on commercially prepared one.

  1. Attempt to prepare anti-human globulin locally.
  2. To compare locally prepared anti-human globulin reagent against commercial ones.
  3. To encourage locally prepared anti-human globulin reagent.
  4. It will create an avenue to teach others how to prepare anti-human globulin reagent locally. Successful attempts at this project will give scientist the zeal to prepare reagents locally and not to depend on the commercial products. The knowledge gotten from this project will help to reduce unemployment since the practice of local preparation of these reagents can serve as source of income.

Polyclonal AHG:– These are antibodies that are secreted by different B cell lineages within the body.

Monoclonal AHG:- They are an artificially produced antibody that neutralizes only one specific antigen (foreign protein).

FAB fragment:- This is known as antigen binding fragment located on the hand of the Y axis of an immunoglobulin. It contains the antigenic determining region (CDR). It composes of both variable and constant region.

FC fragment:- This is known as critalizable fragment, Located on the stalk of the Y axis of immunoglobulin. It composes of the constant region.
